Notes: | The Bcor probe template used in this study by Wamstad & Bardwell, 2007 [PMID:17344103] is described as follows: An amplicon containing nucleotides 280-1294 of the Bcor open reading frame was generated by PCR from a bacterial artificial chromosome containing the mouse Bcor gene using the following primers, forward: 5'-TACGGGTACCATCGATGGCCTGGCAGCACT-3' and reverse: 5'-CATGTCTAGATTGGACAGCCTGGATGTAGAG-3'. The PCR amplicon was gel purified and directly cloned into the Kpn and Xba restriction sites of a pBluescript KS+ vector.
Editors note: although the authors state that the probe contains nucleotides 280-1294 of the Bcor ORF, when these primer sequences are BLASTed against the 3 Bcor mRNA RefSeq sequences: NM_029510.2 (transcript variant a); NM_175044.2 (transcript variant b) and NM_175045.2 (transcript variant c), the resulting start and end points for the PCR amplified fragment are nt280-1260, nt280-1260 and nt466-1446 respectively, the intervening sequence of which is identical for all three. |
